Antigen-antibody Affinity Analysis Service Based on SPR Technology
Antigen-antibody affinity is the binding force between an antibody and an antigenic epitope or antigenic determinant cluster and is essentially a non-covalent force that incorporates attractive forces between amino acids, hydrogen bonding, hydrophobic forces, etc. Antigen-antibody affinity analysis plays an important role in the study of the mechanisms of infectious diseases and related specific drugs, and is a criterion and basis for the selection of antibody drugs. The affinity of an antigen-antibody can be expressed by the affinity constant KD, calculated as KD = [Ab-H]/([Ab][H]) where [] represents the molar concentration, Ab represents the antibody, H represents the semi-antigen and Ab-H represents the binding of the antibody to the semi-antigen. The higher the affinity constant KD, the greater the ability of the antibody to bind the semi-antigen.

The strength of antibody affinity depends on the degree of fit between the antibody counterpart and the epitope of the antigen used, including the size of the contact area, the degree of affinity and the distribution of the dotted groups and hydrophobic genes. Antibody affinity reflects the ability of an antibody molecule to react with a determinant cluster of antigen molecules or semi-antigens and is an important indicator of antibody quality, reflecting the strength of binding between the antibody and the corresponding antigen. The higher the affinity, the higher the degree of binding between the antibody and the corresponding antigen, and is an important guide to the application of antibodies.
Surface plasmon resonance (SPR) in the process of antigen-antibody affinity analysis
The affinity effect of an antigen-antibody is the basis for the screening of antibody drugs and is a key parameter of antibody drugs, which usually affects the function and efficacy of the antibody. In practical development, there are many aspects of the conventional screening of antibodies that require more detailed improvement, including affinity, immunogenicity, half-life, etc. Researchers have done a lot of research and practice in the field of antibodies over the years, with antibody affinity maturation being one of the key areas of research. Theoretically, increasing the affinity of antibodies can help improve the specificity and potency of antibodies, help reduce the dose of drugs and reduce toxic side effects. Furthermore, the development of antibody affinity maturation techniques will not only contribute to the development and quality improvement of antibody drugs, but also to a better understanding of the mechanisms of antibody-target interactions and better understanding of target function. Based on the importance of antigen-antibody affinity studies in antibody drug development, Creative Proteomics is committed to providing you with high-throughput antigen-antibody affinity analysis services through SPR technology.
